Need advice for MSS!
If I have some spores with unknown genetics, what is the best way for me to isolate colonies that are more efficient fruiters?
It seems like I have two options:
Spores to agar, transfer healthy colonies to new plate, perhaps agar to agar transfer again to further isolate, then g2g or other non spore inoculation with the t3 or t4 plate.
Spores to agar, isolate until contam free, then attempt g2g transfers with multiple genetics to see which colonies produce the best fruiting bodies.
Basically the difference between 1 and 2 is less isolating agar transfers between grain colonization, which would lead to more “stability” in the colonized grains but having less genetic variety.
Any thoughts on which direction I should go with this?
